Detection of Contaminated Mycoplasmas on the Cultured Cell Lines


Abstract
Detection of contaminated mycoplasma species on the 82 cultured cell lines was carried out by the direct culture method, the indirect immunofluorescence antibody test, the DNA fluorchrome stain, the immunobloting test and the Mycotect method, and
also
to investigate the influence of antibiotices on the isolation rates of contaminated mycoplasma in the culture media removed antibiotics. The isolated mycoplasmas were identified by the growth inhibition test with specific antisera of standard
mycoplasma
species, and were investigated for their susceptibility against lincomycin and josamycin in vitro.
The mycoplasma contamination rates of tested cell lines were found out 39.0% by the direct culture. 42.6% by the indirect immunofluorescence antibody, 43.9% by the DNA fluorochrome, 37.8% the imunobloting and 0% by the MycoTect method.
The isolation rate of mycoplasmas from cultured cell lnes was increased by the cell lines were subcultured three times in the medium without antibiotics.
The isolated mycoplasmas were identified the 34.4% as M. orale, 25.0% as M.hominis, 12.5% as M. hyorhinis, 9.4% as M. fermentans and M. salivarium. Respectively, but the 9.4% couldn's classified.
The minimum inhibitory concentration ranges for lincomycin and josamycin against to the isolated mycoplasmas were from 0.4 to 12.5¥ìg/ml, respectively.
The infected mycoplasmas on the cell lines could eliminated with the adminstration of josamycin and the specific antisera in the media.
